0.5 % (w/v) locust bean gum (LBG) in 50 mM sodium citrate buffer at pH 4.8

The enzyme preparation (0.2 ml) was added into 1.8 ml of substrate.

At 50 °C in the water bath for 5 min

Glucanase

1 % (w/v) carboxymethylcellulose (CMC) in 50 mM sodium citrate buffer at pH 4.8

The enzyme preparation (1 ml) was added into 1 ml of substrate and 1.0 ml of sodium citrate buffer

At 50 °C in the water bath for 30 min

Glucosidase

15 mM cellobiose in 50 mM sodium citrate buffer at pH 4.8

The enzyme preparation (0.2 ml) was added to 0.2 ml of substrate and 0.2 ml of sodium citrate buffer

At 50 °C in the water bath for 15 min

Fructosyltransferase

40 % (w/v) sucrose in 50 mM sodium acetate buffer at pH 5.4

The enzyme preparation (1.0 ml) was added into 49 ml of substrate

At 50 °C in the water bath for 5 min
